David Artell will take charge of Crewe for the first time when Luton visit Gresty Road.
The former Alex captain has made the step up from the club's academy to replace Steve Davis, who was sacked following last weekend's 3-0 defeat at Mansfield.
Artell has been boosted by the news that Chelsea winger Alex Kiwomya's loan spell has been extended for the rest of the season while the likes of Ben Nugent, Danny Hollands and George Ray are pushing for recalls.
Captain and top scorer Ryan Lowe is fit again after a hamstring injury and also vying for a starting place.
Luton defender Glen Rea is suspended after his late red card in the FA Cup defeat against Accrington Stanley.
Striker Jack Marriott is pushing for a starting berth alongside top-scorer Danny Hylton after bagging a brace in the Checkatrade Trophy victory at Chesterfield on Wednesday night, while new signing Lawson D'Ath could make his debut for the Hatters in midfield.
Town boss Nathan Jones also has a tough decision to make at left-back, with Jack Senior in contention to start after a strong performance in midweek.
The Hatters will be without Alex Gilliead though, after parent club Newcastle sent him on loan to Bradford instead. Midfielder Cameron McGeehan (broken leg) is also out.
