Ayr have included new signing Adam Hunter in their squad to face Stenhousemuir at Somerset Park.
The defender arrived from Rangers on Thursday and is in line for a quickfire debut as Austin McCann and Jonathan Tiffoney are both suspended.
Michael McGowan is expected to miss out with a knee injury although John Robertson is likely to be available.
Manager Mark Roberts said: "We are getting into the real stuff now and we go into it confident after a great 6-1 win over Clyde last weekend.
"Stenhousemuir had a great result last weekend as well when they defeated Brechin 4-0 so we need to ensure that we are playing at the top of our game from the first whistle."
Stenhousemuir will be without suspended striker John Gemmell.
The former Albion Rovers man, who has scored two goals in two games for the Warriors, misses out along with the experienced Eric Paton (hip).
Manager Martyn Corrigan is expecting goals, he said: "We had a great win last weekend over Brechin when we scored four times and Ayr did even better by knocking six past Clyde, so I cannot imagine either of us sitting back and looking for the draw.
"We have had them watched and know they have good football players but we will be going out to pick up our first three points of the season."
