They now travel to play Norwich tomorrow evening.
Mourinho said: “How can they play again in less than 48 hours? The distances they run, the sprints, the intensity… how are we going to control that?
"It’s a crime that they are going to play football again on the 28th.
"It’s against every rule of physiology, biology. But that’s the way it is.”
Spurs’ £65million record buy Tanguy Ndombele is set to start at Norwich after crying off against Brighton.
Jose said afterwards: “I can’t tell you he is injured but I can say yesterday he told me he was not feeling in conditions to play.
"Not based on injury but based on fears of previous injuries he’s had.”
Brighton took the lead through Adam Webster’s 32nd-minute header.
Harry Kane equalised in the 53rd minute and Dele Alli won it on 72 minutes.
Mourinho said: “Another goal and another incredible performance and effort from Dele.”
Albion manager Graham Potter, whose team host Bournemouth tomorrow lunchtime, said: “We’re disappointed but we come away with lots of positives.
"It’s frustrating but we have to take that frustration out on Bournemouth in a few hours’ time. It’s very difficult to play again so soon but we have to.”
